Abstract: background: oral isotretinoin therapy has completely changed the treatment of acne. the clinical adverse effects of isotretinoin are classified into two groups: mucocutaneous and systemic toxic. laboratory alterations such as dyslipidemia and increase of hepatic enzymes are reported. objectives: to evaluate the tolerability of oral isotretinoin with special attention to lipid metabolism, hepatic function, and clinical adverse reactions. methods: 150 patients clinically diagnosed with acne being treated with oral isotretinoin were included in the study. clinical and laboratory evaluations were conducted before the start of therapy, one month after, and every three months thereafter until the completion of treatment. results: of the 150 patients evaluated, 48% were female and 52%, male. they were aged from 15 to 32 years. the most prevalent mucocutaneous adverse reaction associated with isotretinoin therapy was cheilitis, reported by 94% of the patients. systemic clinical effects were much less common. cholesterol, triglyceride, and transaminase levels were assessed and significant changes were not observed during treatment. conclusion: patients showed adverse effects compatible to those described in the literature, which are mostly controlled with symptomatic medication. isotretinoin is a safe drug and adverse and laboratory effects are well tolerated.

Abstract: oligonychus ilicis (mcgregor) (acari) (tetranychidae) is one of the most important pests of coffea canephora pierre & froehner. in this study, the effect of different temperatures in the development of red mite in c. canephora leaves was evaluated. eighty adult females were placed in 40 cages to oviposition containing 4 cm leaf discs at 21, 24, 27, 30 and 33 oc. to evaluated embrionary development two randomly selected eggs were allowed to complete development. after larvae hatch, evaluations were performed each 12 hours to determine the survival and larval period. to evaluate longevity one male was transferred to a cage with one female to mate. the thermal threshold of development and thermic constant were determined to egg adult period of development. the development period of immature stages of o. ilicis decreased as soon as the temperature increased. the stages of egg, larvae, protochrysalidae, protonymph, deutochrysalidae, deutonymph and teleiochrysalidae ranged from 10.4 to 4.3, 2.4 to 1.2, 2.0 to 1.0, 2.3 to 1.2, 1.9 to 1.0, 3.0 to 1.5 and 2.3 to 1.0 days, respectivelly. the temperature increase affected the period of egg adult development and adult longevity the thermal threshold of development was 9.0 oc to egg-adult period and thermal constant was 256.4 gd. at laboratory conditions, temperatures between 24 and 30 oc favours the mite development in c. canephora. the thermal threshold of development does not constrain the o. ilicis occurrence with c. canephora under field conditions in the state of espirito santo, brazil.

Abstract: Long COVID has been studied as different sequelae that some individuals can develop after the acute phase of the disease. Persistent symptoms such as dry cough, fatigue, and dyspnea can remain after six months of COVID-19 cure. Others such as lung fibrosis, kidney injury, and thrombotic risk also are observed. Here, a deep review of each human organ and system infected by the virus was performed aiming to show how molecules expression and cell signaling can induce the organism cure or injuries and, subsequently sequelae. The review also suggests the importance of public health surveillance for these cases including a more comprehensive analysis of molecular biology tools that can clarify and assist in the prognosis, treatment, and preventive methods for potentially more serious disorders in post-COVID-19 patients.

Abstract: in malacological survey conducted at mariana municipality, state of minas gerais, brazil, between april 2003 and february 2004 were found biomphalaria glabrata (say, 1818) and physa marmorata guilding, 1828 mollusks harboring digenea larval trematodes. in b. glabrata were observed, in addition to cercaria of schistosoma mansoni sambon, 1907, strigeocercaria, two types of xiphidiocercaria, ocellifera cercaria and two types of gymnocephalus, while in p. marmorata the only larval form emerging was echinostome cercaria. some of cercariae found differ from those described in the literature.

Abstract: fluorescent in situ hybridization (fish) with commercial probes covering the elastin gene (eln) was used to determine the frequency of the 7q11.23 deletion in 18 children clinically diagnosed with williams-beuren syndrome (wbs). a de novo deletion was detected in 15 of the children (83%). diagnostic investigation for wbs started late in childhood (median = 5.8 years). all the children showed facial features typical of the syndrome, mental retardation and developmental delay. over-friendliness was observed in the majority of cases. clinodactyly of the 5th finger (n = 13), cardiovascular disease (n = 9), loquacity (n = 9), low birthweight (n = 8), and failure to thrive (n = 9) were observed only in those children with the deletion. respiratory problems (n = 9), though not previously reported in the literature, was a common finding in the group studied. our results confirmed that fish is useful in identifying 7q11.23 deletions in cases of wbs. clinical manifestations were more evident in the deletion-positive children.
Effect of sugarcane fiber digestibility, conservation method and concentrate level on the ruminal ecosystem of beef cattle

Abstract: The aim of this study was to investigate the effects of sugarcane neutral detergent fiber digestibility (NDFD), conservation method, and concentrate level on the ruminal microbial population of steers. Eight ruminal-cannulated Nellore steers were distributed in two contemporary 4 × 4 Latin Square design with a 2 × 2 factorial arrangement of treatments. Experiment 1: diets were formulated with 60% of concentrate level, and two sugarcane genotypes (high or low NDFD) either freshly cut or as silage. Experiment 2: diets were formulated with two levels of concentrate (60 or 80%), and two sugarcane genotypes (high or low NDFD) offered as freshly cut. Each experimental period lasted for 14 d, with the last 4 d used for ruminal fluid collection. Three cellulolytic bacteria (Fibrobacter succinogenes, Ruminococcus albus, Ruminococcus flavefaciens), two amylolytic (Streptococcus bovis, Ruminobacter amylophilus), and a lactate fermenting microorganism (Megasphaera elsdenii) were quantified by qPCR. Experiment 1: diets with fresh sugarcane increased the population of S. bovis, and M. elsdenii. Sugarcane with high NDFD increased F. succinogenes population only when sugarcane was offered as freshly cut. Experiment 2: increasing concentrate in the diet decreased S. bovis population, and increased R. amylophilus. Sugarcane with high NDFD increased the population of cellulolytic bacteria only at the 60% concentrate diet. Providing sugarcane with high NDFD favored the growth of fibrolytic bacteria, and this effect were dependent on the conservation method and on diet concentrate level. In addition, sucrose appears to have great effect on the composition of ruminal microflora, especially S. bovis

Abstract: introduction: the autonomic nervous system governs the short and medium term, the necessary modifications to adapt blood pressure and heart rate to changes in posture. objective: to analyze the hemodynamics variables in healthy individuals when subjected to positional gravity. materials and methods: sixty healthy individuals, divided in groups based on age, being gi (n = 20), from 15 to 20 years old, gii (n = 20), from 21 to 40 years old, giii (n = 20) from 41 to 60 years old, being submitted to the position gravity through the gps equipment and they had the cardiac frequency and blood pressure monitored throughout all the time of inversion. results: a significant reduction of heart rate and blood pressure in the rest position when compared after 3, 6, 9 and 12 minutes of inversion, however 3 minutes after return to the orthostatic position this variables came back near to the baseline. about the blood pressure diastolic, it was not observed a significant decrease in the gi and gii group in the rest position when compared after 3 minutes; however in the giii group it was observed a significant drop. after 6 minutes of inversion only the gi group showed a significant drop. when analyzed 9 minutes after the position, only the giii group showed a significant dro. after 12 minutes the inversion, it was also showed a blood pressure diastolic drop only in giii group when compared to the values of the rest position. returning to the vertical position, all groups returned to the baseline values after 3 minutes. conclusion: the gravitational positioning changes the hemodynamics variables causing a drop of the values of the heart rate and the blood pressure during the inversion period.

Abstract: nonmedullary thyroid carcinoma, originating from thyroid epithelial cells, is the most frequent thyroid malignant neoplasia. since 1955, there has been increasing evidence that this cancer may have a familial predisposition. it is now established that around 4.2% of all nonmedullary thyroid carcinomas occurs on the background of familial predisposition. these cases are often more aggressive, due to early onset, multifocality and a higher percentual of recurrences. an autossomal dominant inheritance pattern appears likely in most families, although the exact genes responsible for this syndrome have not yet been identified. patients affected by this cancer should be treated with total thyroidectomy routinely and, in most cases, lymph node dissection, followed by iodine ablation and tsh suppressive therapy with levothyroxine. some authors also recommend that first-degree relatives of patients with nonmedullary thyroid cancer (especially women) should be submitted to neck ultrasound for thyroid cancer screening, aiming early diagnosis for better treatment results.
Improving Kinetic or Thermodynamic Stability of an Azoreductase by Directed Evolution

Abstract: Protein stability arises from a combination of factors which are often difficult to rationalise. Therefore its improvement is better addressed through directed evolution than by rational design approaches. In this study, five rounds of mutagenesis/recombination followed by high-throughput screening (≈10,000 clones) yielded the hit 1B6 showing a 300-fold higher half life at 50°C than that exhibited by the homodimeric wild type PpAzoR azoreductase from Pseudomonas putida MET94. The characterization using fluorescence, calorimetry and light scattering shows that 1B6 has a folded state slightly less stable than the wild type (with lower melting and optimal temperatures) but in contrast is more resistant to irreversible denaturation. The superior kinetic stability of 1B6 variant was therefore related to an increased resistance of the unfolded monomers to aggregation through the introduction of mutations that disturbed hydrophobic patches and increased the surface net charge of the protein. Variants 2A1 and 2A1-Y179H with increased thermodynamic stability (10 to 20°C higher melting temperature than wild type) were also examined showing the distinctive nature of mutations that lead to improved structural robustness: these occur in residues that are mostly involved in strengthening the solvent-exposed loops or the inter-dimer interactions of the folded state.
Factors associated with HIV infection among delivered women in Sergipe, Brazil

Abstract: This was a case-control study, with 39 cases and 117 controls that gave birth at the official health system hospitals. All patients were tested for HIV at hospital admission, using a rapid test and were interviewed about socioeconomic conditions and health attitudes and practices. Univariate and multivariate logistic analysis were performed to evaluate the factors associated with HIV infection.In the univariate analysis, association with HIV positivity was found for the variables "antenatal HIV test" (OR: 4.44; CI: 1.93 – 10.29) and "intravenous drug use" (OR = 12.08; 95% CI 1.28 – 8). Three patients were intravenous drug users, all HIV+. After logistic multivariate regression, not being tested for HIV during antenatal care (OR = 4.98; 95% CI: 2.13–12.22; p < 0.001) and lack of knowledge on how to prevent HIV infection (OR = 2.56; 95%CI: 1.09 – 6.27; p = 0.030) were independently associated with HIV positivity.Drug use, limited knowledge about how to prevent AIDS, and lack of HIV testing during pregnancy were risk factors for infection with HIV. Although it was not conceived to evaluate effectiveness of procedures to prevent vertical transmission, the risk factors here detected may corroborate official recommendation for rapid HIV testing at delivery as an effective procedure to prevent vertical transmission.The AIDS epidemic in Brazil shows a downward trend towards stability since 1996, coinciding with the introduction of antiretroviral therapy. The deceleration of the epidemic however did not occur homogeneously within regions. In the Northeast region for example, the incidence rates of AIDS continue to grow [1].Heterosexual transmission has increased since the early 90s, leading to an increased number of women testing positive for HIV [2,3]. This can be observed by the dramatic change from the 28:1 male-to-female ratio in the beginning of the epidemic to the 1.3:1 ratio of current HIV cases [4].
